感染-免疫-肿瘤

天然免疫是抵抗感染与肿瘤的第一道防线。天然免疫失调引起各种感染性疾病、自身免疫病和肿瘤。识别细胞内DNA的cGAS-STING通路,对抗病毒和抗肿瘤的免疫监视及治疗至关重要。蒋争凡实验室是发现STING蛋白重要功能的三个实验室之一(命名为ERIS),近年对其生理学功能及其调节进行了深入系统的研究:发现STING的二聚体化对其激活天然免疫反应重要(PNAS, 2009);STING-TBK1活化STAT6招募免疫细胞(Cell, 2011);NFkB在cGAS-STING中的重要作用(JI, 2017a);发现3'3'-cGAMP特异性磷酸二酯酶(Cell Research, 2015);不同Caspases负调控天然免疫反应,包括炎症小体活化的Caspase-1(Immunity, 2017;JI, 2017b)及凋亡活化的Caspase(Molecular Cell, 2019)切割天然免疫关键蛋白,避免过度天然免疫及细胞凋亡“免疫沉默”的分子机制;锰离子是细胞内cGAS的第二个激活剂,以独特方式激活cGAS(Immunity, 2018;Cell Reports, 2020),并发明锰佐剂可激活细胞免疫、体液免疫和黏膜免疫(CMI, 2021)及增强肿瘤免疫治疗的“锰免疗法”(Cell Research, 2020);硫酸化糖胺聚糖是STING共配体及STING发生高尔基体转运的原因(Immunity, 2021);STING相分离形成独特立方膜结构“STING相分离器”调节天然免疫(Nature Cell Biology, 2021)。此外,发现PCBPs-AIP4介导的蛋白质降解途径负调控天然免疫关键分子MAVS降低或避免RNA病毒感染引发的过度反应(Nature Immunology, 2009; Cell Research, 2012)以及NEMO-IKKα/β对于RIG-I-MAVS通路中激活TBK1/IKKε至关重要(PLoS Pathogens, 2017)。
